Protocols
The 24sure protocol comprises 5 distinct stages. Workflow is typically organised around 8 samples though the option to complete some stages in microtitre plates can scale this number to 96 samples and beyond.
Amplification. SurePlex is used to extract DNA from single cells supplied in PBS, fresh or frozen. Following a quality control, amplified template may be used immediately or stored until required. Duration 2 hours.
Labelling. Amplified template is labelled using random primers and Cy dyes. Volumes are reduced by precipitation or plate evaporation. Duration 3 hours.
Hybridization. Labelled fragments are hybridized to 24sure arrays and washed. Duration 3 hours.
Scanning. A dual channel fluorescent laser scanner is used to create TIFF images of arrays. Duration up to 30 minutes depending upon scanner performance.
Data analysis. 24sure software is used to generate results for each sample. Analysis is fully automated and results are straightforward to interpret. Duration 10 minutes.
